Cell rupture microfluidic device using nano needle allay for damagefree extraction of organelles

Abstract

In order to realize advanced cell function analysis, soft cell rupture device using glass nano needle arrays is developed for damage-free extraction of organelles. The cell rupture device has a simple pneumatic PDMS microvalve with nano needles fabricated on a glass substrate. By applying the pneumatic pressure, the introduced cells are pushed to the nano needles and the cell membrane is mechanically broken with small pressure. Cytoplasnic matrix (cytosol), mitochondria and lysosome trapped in the cytoskeleton are efficiently extracted within 1 sec with this device.
