Abstract

In this paper, we developed a high performance PDMS microfluidic device for sequential trapping, labeling and content extraction from single cells. The high performance single cell manipulation device consists of a main channel, single cell capture pockets, drain channels, and single cell lysate collection channels. The spontaneous single cell lysate extraction in the single cell pocket with chemical solution of 10% sodium dodecyl sulfate (SDS) was achieved. Since this device demonstrated the successive single cell isolation, labeling and single-cell lysate extraction, the pretreatment of the capillary electrophoresis can be completed.
