Abstract

We present a novel peptide-based ligand screening system for G protein-coupled receptors (GPCRs) in water-in-oil (W/O) microdroplets. This system is an in vitro gene screening system for peptides that activate a target GPCR expressed in yeast. We show that the system is applicable to human GPCRs. This system will be a powerful tool for identifying novel peptide ligands for liganded mammalian GPCRs and orphan mammalian GPCRs.
