N2 - We have studied prototypes of platelet substitutes and focused on the dodecapeptide (H12); a part of the amino acid sequence of fibrinogen as activated platelets, and conjugated H12 to the surface of polymerized albumin particles (polyAlb) as biocompatible and biodegradable particles. The H12-conjugated polyAlb (H12-polyAlb) adding to the thrombocytopenic-imitation blood enhanced the platelet thrombus formation on a collagen-plate in vitro. We confirmed that H12-polyAlb had a hemostatic effect, based on the analysis of the tail bleeding time of thrombocytopenic rats. H12-polyAlb is a suitable candidate for an alternative to human platelet concentrates infused into thrombocytopenic patients.
