Improved passive cell distributing method for micro cellular diagnostic well array

We developed a novel biological cell injection method for a large number of diagnostic wells fabricated in parallel. This method facilitates simultaneous cell injection into a microwell array with a simple channel configuration, which can distribute cells evenly to each microwell only by introducing cell suspensions from the inlet. In order to enhance the uniformity of the numbers of injected cells in each microwell, we developed the new concept of flow interchange, which can suppress deviation of the cell flow profile in the cell loading channels. The uniformity of the injected fluorescent beads in each microwell was evaluated using prototype devices of eight microwell arrays. It was confirmed that the flow interchange structure is effective to realize uniform injection. The coefficient of variation of bead numbers in eight microwells was 20.7%.
