The role of phospho-protein in bone formation was investigated in this study. A phosvitin layer was formed on a collagen sheet by soaking the sheet in a fluid containing phosvitin of 0.04wt% and various concentrations of dimethylsuberimidate. The dimethylsuberimidate concentration was changed from 0.04 to 0.3wt% to find the appropriate concentration for phosvitin-collagen layer formation. The composite layer was soaked in physiological solutions and the nucleation process of calcium phosphates was observed by using atomic force microscopy (AFM). It was found that small hydroxyapatite (HAP) crystals were formed throughout the layer with their a-axes arranged perpendicular to it. On the other hand, HAP with its c-axis perpendicular to the layer was formed when phosvitin was not present. This makes it clear that the roles of phosvitin are to enhance nucleation of HAP, and to control the orientation of HAP crystals on collagen in the same way as that observed in human bone.
